dc.description.abstractการศึกษาเรื่องการพัฒนาชุดตรวจสอบสารกลุ่มไนโตรฟูแรนในอาหารสัตว์ ด้วยวิธีคัลเลอริเมตริก พบว่า การใช้ไดเมทิลฟอร์มาไมด์ ร่วมกับ 1 โมลาร์ เอทาโนลิค โพแทสเซียมไฮดรอกไซด์ ที่อัตราส่วน 10:1 จะทำให้สารกลุ่มไนโตรฟูแรนเกิดการเปลี่ยนแปลงสีได้ดีที่สุด ซึ่งจะได้สารมีสีที่มองเห็นได้ด้วยตาเปล่า ดังนี้ ไนโตรฟูราโซน จะเกิดสีม่วงแดง, ไนโตรฟูแรนโตอิน จะเกิดสีน้ำตาล, ฟูราโซลิโดน จะเกิดสีม่วง และ ฟูราลตาโดน จะเกิดสีน้ำเงิน ซึ่งสามารถหาปริมาณสารดังกล่าวได้โดยการเทียบความเข้มของสีที่มองเห็นกับแถบสีมาตรฐาน โดยมีช่วงการวิเคราะห์ที่เหมาะสม คือ 0.5 ถึง 50 ppm เมื่อทำการตรวจสอบความใช้ได้ (Validation) ของชุดตรวจสอบที่พัฒนาขึ้นกับตัวอย่างอาหารสัตว์ชนิดต่างๆ ที่ทำการเติม (spike) สารกลุ่มไนโตรฟูแรน 50 ppm ลงไปในตัวอย่างพบว่าตัวอย่างทุกชนิดเกิดการเปลี่ยนแปลงสี โดยสีที่เกิดขึ้นเป็นสีเดียวกับแถบสีมาตรฐาน ผลการวิเคราะห์ความใช้ได้ของชุดตรวจสอบโดยใช้ UV-Visible สเปกโตรโฟโตมิเตอร์ พบว่ามีความแม่นยำ (RSD) อยู่ในช่วง 0.11 ถึง 0.32%และความถูกต้อง (% Recovery) อยู่ในช่วง 81 ถึง 98% และโดยใช้ HPLC พบว่ามีความแม่นยำ (RSD) อยู่ในช่วง 1.97 ถึง 3.51 % และความถูกต้อง (% Recovery) อยู่ในช่วง 73 ถึง 93 % สำหรับผลการหาผลบวกลวง (false positive) ที่อาจเกิดขึ้นกับชุดตรวจสอบ พบว่าชุดตรวจสอบที่พัฒนาขึ้นจะไม่ให้ผลบวกลวงใดๆกับตัวอย่าง และยาปฏิชีวนะอื่นๆ คือ คลอแรมฟินิคอล, นอร์ฟล๊อกซาซิน และ ฟลูมิควิน-
dc.description.abstractalternativeThe test kit of Nitrofurans was developed based on colorimetric method. This test kit was suitable for testing Nitrofurans antibiotics used in animal feed and livestock drugs. The test kit gave a specific color with specific type of nitrofurans. The combination of Dimethylformamide (DMF) and 1M Ethanolic Potassium hydroxide (1M KOH/EtOH) at the ratio of 10:1 gave the best results in the aspect of color and stability. The color could be visualized clearly in these following consequences: Nitrofurazone, Nitrofurantoin, Furazolidone and Furaltadone as purple, brownish, violet and blue color, respectively. The quantitative analysis could be achieved by using standard color band strip. The concentration of nitrofurans from 0.5-50 ppm could be suitably detected by this test kit. The method validation of this test kit was carried out by using UV-Visible Spectrophotometer and HPLC. These gave the recovery and precision in a range of 81-98% 0.11-0.32 % RSD for UV-Vis and in a range of 73-93% and 1.97-3.51 % RSD for HPLC. Finally, this test kit did not give any false positive results to other samples and antibiotics such as Chloramphenicol, Norfloxacin and Flumiquin.-
