dc.description.abstractการศึกษาเกี่ยวกับปัญหายาเสพติดให้โทษทางหนึ่ง คือ การพัฒนาวิธีการวิเคราะห์ยาเสพติดและเมตาโบไลท์ของยาเหล่านั้นในของเหลวจากร่างกายด้วยวิธีที่มีความถูกต้องเชื่อถือได้และมีความไวสูง วิธีเอนไซม์มัลติไพลด์อิมมิวโนแอสเสย์เป็นวิธีหนึ่งที่สะดวกและรวดเร็วไม่ต้องใช้เครื่องมือที่มีราคาแพง แต่ขณะนี้ยังต้องซื้อน้ำยาสำเร็จรูปซึ่งมีราคาแพงจากต่างประเทศ การวิจัยครั้งนี้ได้พัฒนาน้ำยาสำเร็จรูปดังกล่าวบางอย่าง คือ สารละลายแขวนลอยไมโครคอคคัสลูเทียส มอร์ฟีน-ไลโซไซม์คอนจูเกต สำหรับใช้ในการวิเคราะห์อนุพันธ์มอร์ฟีนในปัสสาวะ วิธีเอนไซม์มัลติไพลด์อิมมิวโนแอสเสย์ที่พัฒนาขึ้นโดยใช้น้ำยาที่เตรียมได้มีความไว 180 นาโนกรัม/มิลลิลิตรปัสสาวะ มีความถูกต้องและแม่นยำพอใช้ สามารถวัดได้วันละ 100 – 150 ตัวอย่าง โดยใช้เวลาในการวัดต่อตัวอย่าง 1-2 นาที การวิเคราะห์อนุพันธ์มอร์ฟีนในปัสสาวะโดยวิธีเอนไซม์มัลติไพลด์อิมมิวโนแอสเสย์โดยใช้น้ำยาที่เตรียมขึ้นเองสามารถลดค่าใช้จ่ายได้ประมาณ 60 เท่าของเมื่อใช้น้ำยาสำเร็จรูปจากต่างประเทศ และลดค่าใช้จ่ายได้ประมาณ 10 เท่าของเมื่อใช้วิธีเรดิโออิมมิวโนแอสเสย์ แม้ว่าวิธีเอนไซม์มัลติไพลด์อิมมิวโนแอสเสย์จะมีความไวต่ำกว่าวิธีเรดิโออิมมิวโนแอสเสย์ แต่ก็มีความไวพอที่จะใช้วัดปริมาณอนุพันธ์มอร์ฟีนในปัสสาวะได้ และมีความถูกต้อง ความแม่นยำใกล้เคียงกับวิธีเรดิโออิมมิวโนแอสเสย์ ปริมาณอนุพันธ์มอร์ฟีนในปัสสาวะชาวไทยภูเขาที่ใช้และติดฝิ่นจำนวน 60 ตัวอย่างที่วัดโดยวิธีทั้งสองมีความสัมพันธ์กันและมีค่าสัมประสิทธิ์สหสัมพันธ์เท่ากับ 0.94 ผลจากการศึกษาครั้งนี้ยังเป็นแนวทางสำหรับการพัฒนาน้ำยาสำเร็จรูปราคาถูกสำหรับวิธีเอนไซม์มัลติไพลด์อิมมิวโนแอสเสย์ที่ใช้วิเคราะห์ยาเสพติดและสารประเภทอื่น ๆ ด้วย
dc.description.abstractalternativeOne of the essential steps to solve the problem of narcotic drugs is the development of detection method of these drugs and their metabolites in body fluids. The method should be quantitatively reliable and sensitive. Enzyme multiplied immunoassay technique was a rapid and convenient technique which does not require complicate and expensive equipments. However the reagent kit is imported at high cost. This thesis describe the preparation of Micrococcusluteus suspension and morphine-lysozyme conjugates for determining the morphine derivatives in urine. Using the author’s own preparations, the sensitivity of enzyme multiplied immunoassay technique was 180 ng/ml of urine. The accuracy and precision of the method were acceptable. This method could assay 100-150 samples per day and each sample took only 1-2 min. comparing this method with radio-immunoassay, it was found that the prepared enzyme multiplied immunoassay technique kit was 60 and 10 times less expensive than that of the imported enzyme multiplied immunoassay technique kit and radioimmunoassay kit respectively. Eventhough enzyme multiplied immunoassay technique was less sensitive than the radioimmunoassay technique but the accuracy and precision of both methods were comparable. Study in 60 urine samples of opium-addicted hill tribes of Thailand showed high correlation of enzyme multiplied immunoassay technique and radioimmunoassay with correlation coefficient of 0.94. This study can serve as a potential application for preparing inexpensive enzyme multiplied immunoassay technique kit for some other drugs or substances.
