| Abstract: | เชื้อรา Absidia sp. BA16 สามารถแปรรูปกรดลิโทโคลิกไปเป็นอนุพันธ์กรดลิโทโคลิกหลัก 2 ชนิด ซึ่งมีค่า Rf และสีที่ปรากฏขึ้นบนแผ่นโครมาโตกราฟแบบผิวบางใกล้เคียงกับกรดอูโชดีออกชีโคลิกและกรดคีโนดีออกชีโคลิก สกัดแยกอนุพันธ์กรดลิโทโคลิกออกจากอาหารเลี้ยงเชื้อด้วยเอทธิลอชีเตทและทำให้บริสุทธิ์ โดยใช้ซิลิกาเจลคอลัมน์โครมาโตกราฟี หลังจากนั้นตกผลึกด้วยสารผสมเอทธิลอชีเตทและเฮกเชน ได้ศึกษาโครงสร้างของอนุพันธ์กรดลิโทโคลิกโดยการวิเคราะห์ธาตุองค์ประกอบอินฟราเรสเปคตรัม คาร์บอน-13 และโปรตอนนิวเคลียร์แมกเนติกเรโชแนนซ์ พบว่า อนุพันธ์กรดลิโทโคลิกนี้เป็นกรด 3 แอลฟา 15 เบตาไดไฮดรอกซี 5 เมตาโคลานิก |
| Other Abstract: | The fungus Absidia sp. BA16 was found to be able to transform lithocholic acid into two major derivatives which possessed the Rf value and the color after developed on TLC plate are similar to those of ursodeoxycholic acid and chenodeoxycholic acid. The product was extracted from the culture broth with ethyl acetate and purified by silica gel column chromatography followed by crystallizing with the mixture of ethyl acetate and hexane. The product was identified as 3α, 15β-dihydroxy-5 β-cholanic acid on the basis of elemental analysis, IR, C13-NMR, H1-NMR etc.. |
