dc.description.abstractการวิจัยครั้งนี้มีวัตถุประสงค์เพื่อเปรียบเทียบผลของจีเอ็นอาร์เอซอนาลอกส์ 3 ชนิด คือ D-Ala6 GnRH, D-Ala6 Pro9 ethylamide GnRH และ D-Ala6 Pro9diethylamide GnRH ในการกระตุ้นการหลั่งแอลเอชจากต่อมใต้สมองส่วนหน้าของหนูแรทเพศเมีย อายุ 90 - 120 วัน โดยใช้หนูแรทโดยตัดรังไข่ออกทั้งสองข้างเป็นเวลานาน 1 เดือน แล้วฉีดเอสตรา-ไดออล เบนโซเอทขนาด 50 ไมโครกรัมและโปรเจสเตอโรนขนาด 25- มิลลิกรมต่อตัว เข้าทางใต้ผิวหนังแล้วทิ้งไว้ 72. ชั่วโมง แล้วแบ่งหนูทดลองออกเป็น 4 กลุ่มๆ ละ 10 ตัว คือ กลุ่มควบคุมที่ฉีดน้ำเกลือ กลุ่มฉีด D-Ala6 GnRH, D-Ala6 Pro9 ethylamide GnRH และ D-Ala6 Pro9diethylamide GnRH ขนาด 100 นาโนกรัมต่อตัวเข้าทางเส้นเลือดที่หาง แล้วเก็บเลือดโดยวิธี Cardiac puncture หลังฉีดสาร 15 นาที จากนั้นตรวจวัดปริมาณแอลเอชในซีรัมด้วยวิธีไบโอแอสเสย์ พบว่าระดับแอลเอชในซีรัมหลังจากฉีดน้ำเกลือ D-Ala6 GnRH, D-Ala6 Pro9 ethylamide GnRH และ D-Ala6 Pro9diethylamide GnRH เท่ากับ 300 ± 100, 620±l64, 740 ±134 และ343 ± 113 นาโนกรัมต่อมิลลิลิตรตามลำดับ จากการทดลองนี้แสดงให้เห็นว่าจีเอ็นอาร์เอชอนาลอกส์ชนิด D-Ala6 ที่มีหมู่ ethylamide ในตำแหน่งที่ 10 มีความสามารถในการกระตุ้นการหลั่งแอลเอชมากกว่าจีเอ็นอาร์เอชอนาลอกส์ชนิด D-Ala6 GnRH และ D-Ala6 Pro9 ethylamide GnRH อย่างมีนัยทางสำคัญที่ระดับความเชื่อมั่น 0.05-
dc.description.abstractalternativeThe aim of this study was to elucidate the effect of GnRH analogs; D-Ala6 GnRH, D-Ala6 Pro9 ethylamide GnRH and D-Ala6 Pro9 diethylamide GnRH on LH secretion from anterior pituitary gland of female rats, aged 90-120 days. After one month of ovariectomy, rats were subcutaneously injected with estradiol benzoate (50 ug/rat) and progesterone (25 mg/rat) for 72 hours before the injection of GnRH analogs. The rats were divided into 4 groups (10 rats/group); control group was intravenously injected with saline other 3 groups were injected with D-Ala6 GnRH, D-Ala6 Pro9 ethylamide GnRH and D-Ala6 Pro9 diethylamide GnRH at doses of 100 ng/rat. The bloodSiamples were collected by cardiac puncture 15 minutes after injections. Serum was analyzed I for LH level by bioassay method. It has been found that LH levels after saline, D-Ala6 GnRH, D-Ala6 Pro9 ethylamide GnRH and D-Ala6 Pro9 diethylamide GnRH injection were 300±100, 620±164, 740±134 and 343±113 ng/ml respectively. Results obtained indicated that D-Ala6 GnRH analog with ethylamide at the tenth position is the most effective analog in stimulating LH secretion compared with D-Ala6 GnRH and D-Ala6 Pro9 diethylamide GnRH.-
