dc.description.abstractการศึกษานี้มีวัตถุประสงค์ เปรียบเทียบวิธีมาตรฐานและวิธีรวดเร็ว 3 วิธี ได้แก่ Fluorocult R LMX Broth (LMX) Petrifilm TM E. coli Count Plates (PEC) และ Chromocult R Coliform Agar (CCA) เพื่อการแจงนับ Total Coliforms และ Escherichia coli ในกุ้งกุลาดำแช่แข็ง และนำผลการวิจัยมาประเมินการใช้วิธีรวดเร็ว การแจงนับ Total Coliforms และ E.coli ในกุ้งกุลาดำแช่แข็งทั้งจากที่มีจุลินทรีย์ปนเปื้อนโดยธรรมชาติ 18 ตัวอย่างและจากที่ทำการปนเปื้อน Coliforms และ E.coli ใน 3 ระดับ (10 1, 10 2, 10 3 CFU/g) จำนวน 174 ตัวอย่าง พบว่าค่าการแจงนับ Total Coliforms ที่ระดับการปนเปื้อน <10 CFU/g LMX = MPN>CCA = PEC และค่าการแจงนับ E.coli LMX = MPN = CCA = PEC ความไวในการแจงนับ Total Coliforms และ E.coli LMX = MPN>CCA = PEC ความจำเพาะในการแจงนับ Total Coliforms และ E.coli LMS = MPN = CCA = PEC การเกิด ผลบวกเท็จจากการทดสอบยืนยัน Coliforms LMX>MPN = CCA = PEC การเกิดผลบวกเท็จจากการทดสอบยืนยัน E.coli MPN = LMX>PEC = CCA เวลาที่ใช้ MPN>LMX = PEC = CCA ค่าใช้จ่าย MPN>PEC>CCA = LMX แรงงานที่ใช้ MPN>LMX = CCA>PEC ความสะดวก PEC>CCA = LMX>MPN ความสอดคล้องของวิธีรวดเร็วทั้ง 3 วิธีคือ LMX PEC และ CCA เมื่อเทียบกับวิธีมาตรฐานในการแจงนับ Total Coliforms เท่ากับ 94.2%, 79.2%, 83.2% ตามลำดับ ขณะที่ในการแจงนับ E.coli เท่ากับ 99.2%, 95.8%, 83.2% ตามลำดับ วิธีรวดเร็ว Fluorocult R LMX Broth (LMX) หรือ Petrifilm TM E.coli Count Plates (PEC) หรือ Chromocult R Coliform Agar (CCA) เป็นอีกทางเลือกหนึ่งในการทดแทนวิธีมาตรฐานในการแจงนับ Total Coliforms และ E.coli ในกุ้งกุลาดำแช่แข็ง อย่างไรก็ดีการจะเลือกใช้วิธีหนึ่งวิธีใดขึ้นอยู่กับเป้าหมายของการทดสอบของแต่ละห้องปฏิบัติการในการประเมินข้อดีข้อจำกัดและปัญหาซึ่งขึ้นกับสถานการณ์แวดล้อมของแต่ละห้องปฏิบัติการอันจะก่อให้เกิดประโยชน์สูงสุดen
dc.description.abstractalternativeThe purposes of this study are to compare the conventional to the rapid methods including the Fluorocult R LMX Broth (LMX), Petrifilm TM E.coli Count Plates (PEC), and Chromocult R Coliform Agar (CCA) for the enumeration of Total Coliforms and Escherichia coli in frozen black tiger shrimp in order to evaluate the one of these methods. The enumeration of Total Coliforms and E.coli from 18 samples of regular forzen black tiger shrimp and 174 samples of Coliforms plus E.coli contaminated at three different levels (10 1, 10 2, 10 3 CFU/g) revealed that at the level of<10 CFU/g the Total Coliforms enumeration from LMX = MPN>CCA = PEC and the E.coli LMX = MPN = CCA = PEC. The rapid tests showed the following parameters: sensitivity; LMX = MPN>CCA = PEC, specificity; LMX = MPN = CCA = PEC, false positives for Coliforms; LMX>MPN = CCA = PEC, false positives for E.coli; MPN = LMX>PEC = CCA, duration; MPN>LMX = PEC = CCA, expenses; MPN>PEC>CCA = LMX, labor; MPN>LMX = CCA>PEC, convenience; PEC>CCA = LMX>MPN. Agreement (%) to the conventional method; Total Coliforms; LMX (94.2%), PEC (79.2%), CCA (83.2%); E.coli; LMX (99.2%), PEC (95.8%), CCA (83.2%) were observed. In conclusion, the rapid methods Fluorocult R LMX Broth (LMX), Petrifilm TM E.coli Count Plates (PEC) and Chromocult R Coliform Agar (CCA) are considered as alternatives to the conventional method for the enumeration of Total Coliforms and E.coli in frozen black tiger shrimp. Selecting the suitable method, for the most benefit, however depends on the test and the limitation of the laboratory involved.en
