Abstract:
สาร GABA เป็นผลิตผลจากการ metabolismของ glutamic acid โดยเอนไซม์ GAD ( glutamic acid decarboxylase ) ในงานวิจัยนี้ศึกษาสภาวะที่เหมาะสมในการผลิตข้าวกล้องเพาะงอกให้ได้ปริมาณสาร GABA สูงโดยใช้วิธีการเติม glutamic acid โดยในขั้นแรกเป็นการศึกษาผลของความเข้มข้นของ glutamic acid ต่อกระบวนการงอกและลักษณะปรากฏของข้าวกล้อง โดยแปรความเข้มข้นของ glutamic acid ในช่วง 5-70 mM จากการทดลองพบว่าที่ความเข้มข้น 5-30 mM ไม่มีผลต่อเปอร์เซ็นการงอกและลักษณะปรากฏของข้าว ที่ความเข้มข้น 35-45 mM ไม่มีผลเปอร์เซ็นการงอกแต่ข้าวกล้องงอกที่ได้จะมีลักษณะเปื่อยเล็กน้อย และที่ความเข้มข้นมากกว่า 45 mM เปอร์เซ็นการงอกของข้าวกล้องจะลดลง ข้าวที่ได้จะเปื่อย จากผลการศึกษาในตอนต้นนำมาศึกษาร่วมกับปัจจัยทางกายภาพที่มีผลต่อการผลิต GABA ในข้าวปทุมธานี 1 และ ข้าวเหนียว กข 6 ได้แก่ pH และอุณหภูมิ โดยแปร pH 3 ระดับ คือ 4, 5 และ6 , อุณหภูมิ แปร 3 ระดับคือ 30, 40 และ 50 องศาเซลเซียส และความเข้มข้นของ glutamic acid แปร 3 ระดับคือ 5, 15 และ 25 mM หาปริมาณสาร GABA ในข้าวกล้องเพาะงอกที่ได้ และวิเคราะห์ผลด้วยวิธี response surface เพื่อหาสภาวะที่เหมาะสมในการเพาะงอกข้าวกล้อให้ได้ปริมาณสารGABA มากที่สุดด้วยวิธีการเติม glutamic acid พบว่าภาวะที่เหมาะสมในการเพาะงอกข้าวปทุมธานี1 และ ข้าว กข. 6 คือ pH 5, อุณหภูมิ 40 องศาเซลเซียส และความเข้มข้นของ glutamic acid 15 mM โดยมีปริมาณสาร GABA เท่ากับ 155.14±8.01 และ 154.34±30.63 mg/100g ข้าวกล้องงอก (dry basis) ตามลำดับ ปริมาณ เชื้อทั้งหมด ของข้าวปทุมธานี 1 และ ข้าว กข. 6 เท่ากับ 6.78±0.45 log CFU/g และ 6.15±0.34 log CFU/g ตามลำดับ และ yeast และ mold ของข้าวปทุมธานี 1 และ ข้าว กข. 6 เท่ากับ 97.65±9.35 CFU/g และ 102.38±12.77 CFU/g ตามลำดับ
