| dc.description.abstract | การศึกษานี้มีวัตถุประสงค์เพื่อสังเกตผลของการฝังจี เอ็น อาร์ เอช อะโกนิสต์ ชนิดเดสโลรีลิน ใต้ผิวหลังต่อระบบสืบพันธุ์ของสุนัขเพศเมียก่อนวัยเจริญพันธุ์ โดยใช้ลูกสุนัขเพศเมียสุขภาพแข็งแรงที่เกิดจากพ่อแม่เดียวกัน 6 ครอก ครอกละ 3 ตัว ทำการสุ่มแบ่งลูกสุนัขในแต่ละครอกออกเป็น สุนัขตัวที่หนึ่งและสองซึ่งเป็นกลุ่มทดลอง ได้รับการฝังเดสโลรีลิน ขนาด 10 มิลลิกรัมที่อายุ 4 และ 7 เดือนตามลำดับ และสุนัขตัวที่สามในครอกเดียวกันได้รับการฝังยาหลอกเป็นกลุ่มควบคุม โดยไม่พบความผิดปกติใดๆ เมื่อทำการตรวจสุขภาพสุนัขและบริเวณตำแหน่งของการฝังเดสโลรีลิน หลังจากการฝังจี เอ็น อาร์ เอช อะโกนิสต์สุนัขทั้งหมดได้รับการตรวจระยะการเป็นสัด โดยประเมินจากการเปลี่ยนแปลงทางกายภาพของอวัยวะสืบพันธุ์ภายนอก การเปลี่ยนแปลงของระดับฮอร์โมนโปรเจสเตอโรน และการเปลี่ยนแปลงของเซลล์เยื่อบุช่องคลอด ตามช่วงระยะเวลาที่กำหนดคือ หลังจากการฝังฮอร์โมนหรือยาหลอก 2 ครั้งต่อสัปดาห์เป็นระยะเวลา 1 เดือน หลังจากนั้นจึงทำการตรวจระยะการเป็นสัดทุก 2 สัปดาห์จนสุนัขทุกตัวมีอายุ 13 เดือน เมื่อสิ้นสุดการทดลองทำการวัดขนาดของอวัยวะสืบพันธุ์ภายนอกของสุนัขกลุ่มทดลองและกลุ่มควบคุม ทำการเก็บรังไข่และมดลูกเพื่อตรวจหาการเปลี่ยนแปลงทางจุลกายวิภาค ผลการศึกษาพบว่าสุนัขกลุ่มทดลองที่ทำการฝังเดสโลรีลินที่อายุ 4 เดือนทุกตัว (6/6) อยู่ในระยะการเป็นสัดแบบ แอนเอสตรัส ตลอดระยะเวลาการศึกษา (36 สัปดาห์) ในขรธสุนัขที่ทำการฝังเดสโลรีลินที่อายุ 7 เดือนทุกตัว (6/6) แสดงอาการเป็นสัดภายหลังจากการฝังฮอร์โมน 4 ถึง 14 วัน ในกลุ่มควบคุมพบว่าสุนัข 5 ใน 6 ตัวแสดงการเป็นสัดเป็นสัดในช่วง 1 ถึง 4 เดือนภายหลังจากการฝังยาหลอก ช่วงห่างของระยะโปรเอสตรัสและเอสตรัสของสุนัขกลุ่มทดลองที่ทำการฝังฮอร์โมนที่อายุ 7 เดือนและยาหลอกไม่มีความแตกต่างกัน ในขณะที่ช่วงห่างของอายุไดเอสตรัสของสุนัขที่ทำการฝังฮอร์โมนที่อายุ 7 เดือนมีระยะเวลาน้อยกว่าในสุนัขกลุ่มควบคุม (31±0 วันและ 57.6±15.5 วันตามลำดับ) หลังจากสิ้นสุดการทดลองพบว่า ไม่มีความแตกต่างกันอย่างมีนัยสำคัญของขนาดของอวัยวะสืบพันธุ์ภายนอกของสุนัขกลุ่มทดลองและกลุ่มควบคุม (p>0.05) ผลทางจุลกายวิภาคไม่พบพยาธิสภาพของรังขาและมดลูกทั้งในสุนัขกลุ่มทดลองและกลุ่มควบคุม จึงสรุปได้ว่าการฝังจี เป็น อาร์ เอช อะโกนิสต์ เข้าใต้ผิวหนังของสุนัขเพศเมียที่อายุ 4 เดือนจะไม่แสดงผลในการเหนี่ยวนำการเป็นสัดดังเช่นที่พบในการฝังที่อายุ 7 เดือน และไม่พบความผิดปกติจากการฝังเดสโลรีลินทั้งอวัยวะสืบพันธุ์ภายนอกและภายใน นอกจากนี้การฝังเดสโลรีลินในสุนัขที่อายุ 4 เดือนจะทำให้สุนัขถึงวัยเจริญพันธุ์ได้ช้าลง | |
| dc.description.abstractalternative | The study aimed to investigate effect of GnRH agonist (deslorelin ) implantation on reproductive status of prepubertal female dogs. The placeb o controlled trial study was conducted using eighteen female pups, clinically healthy, from 6 litters, and three litter-mate dogs were randomly selected from each litter. First and second pups were subcutaneously implanted with 10 mg deslorelin and placebo at 4 months of age, respectively, while the third pup was implanted with 10 mg deslorelin at 7 months old. No local or systemic clinical signs from all dogs were observed after implantation. Following deslorelin implantation, oestrous signs (e.g. vulvar swelling, and bloody vaginal discharge) vaginal cytology and serum progesterone evaluated, twice a week for 4 weeks, and then every 2 weeks until all dogs were 13 months old. Vulvar dimensions of all dogs were also examined at 13 months old. At 13 months of age, ovaries and uteri were collected and examined following the ovariohysterectomy. The results demonstrated that the deslorelin implanted pups at 4 month of age did not show induced oestrus (6/6) during 36 weeks of experimental period, while the deslorelin implanted pups at 7 month of age exhibited oestrus (6/6). In the control group, oestrus signs were observed in 5 dogs (5/6) within 2.5 to 7 months after placebo implantation. Proestrous and oestrous intervals were not different between the 7 month-old deslorelin implanted and control groups, whilst dioestrous interval of the 7 month-old deslorelin implanted dogs was shorter (31±0 days) than that of the control dogs (57.6±15.5). No significant difference of vulvar dimensions was observed (P>O. 05). Histological examination revealed there was no inflammatory response of the ovary or uterus in any dog, from either experimental or control group. The study concluded that GnRH agonist implantation failed to induce oestrus in female dogs at 4 months of age, as did in dogs at 7 months old and did not affect the external and internal reproductive organs. The deslorelin implantation can postpone puberty in female dogs at 4 months old. | |
