dc.description.abstract	ในปีพศ.2544-2548 พบว่ามีการระบาดของไวรัสตับอักเสบ เอ เกิดขึ้นหลายครั้งในประเทศไทย ดังนั้นในการศึกษานี้ จึงทำการจำแนกลักษณะทางพันธุกรรมของการระบาดที่เกิดขึ้นใน จ.สุพรรณบุรี จ.สงขลา จ.เชียงราย และ จ.ลำปาง ที่เกิดขึ้นในปีพ.ศ. 2544 และ 2548 นอกจากนั้น ยังหาความสัมพันธ์ของนิวคลีโอไทด์ที่ใช้ในการศึกษาครั้งนี้กับนิวคลีโอไทด์ที่มีรายงานในธนาคารรหัสพันธุกรรม (GenBank) และ ถอดรหัสพันธุกรรมทั้งจีโนมของตัวอย่างที่เลือกมา 1ตัวอย่าง ตัวอย่าง ซีรัมที่ได้รับมาจากการระบาดของ 4 จังหวัด ประกอบไปด้วย จ.สุพรรณบุรี (83 ตัวอย่าง) จ.สงขลา (79 ตัวอย่าง) จ.เชียงราย (79 ตัวอย่าง) และ จ.ลำปาง (59 ตัวอย่าง) เมื่อทำการตรวจหา Anti-HAV IgM โดยวิธี ELISA พบว่าอัตราของผู้ติดเชื้อที่ให้ผลบวกต่อ Anti-HAV IgM รวมทั้งสิ้น 135 ตัวอย่าง ซึ่งคิดเป็น 45% โดยแบ่งเป็น จ.สุพรรณบุรี 40 ตัวอย่าง (48.19%), จ.สงขลา 38 ตัวอย่าง (47.5%), จ.เชียงราย 25 ตัวอย่าง (40.98%) และ จ.ลำปาง 32 ตัวอย่าง (54.24%) จากนั้นนำตัวอย่างที่ได้มาทำการตรวจสอบเพิ่มปริมาณสารพันธุกรรมของเชื้อไวรัสด้วยวิธี RT-PCR โดยใช้ primer ในส่วนของ VP1-P2A junction ซึ่งเป็นบริเวณที่มีความแปรปรวนของรหัสพันธุกรรมสูง จากการจำแนกจีโนไทป์ของเชื้อไวรัสตับอักเสบ เอ ที่เกิดขึ้นในประเทศไทย โดยใช้รหัสพันธุกรรมบริเวณส่วนต่อของยีน VP1-P2A ขนาด 168 bp จากการวิเคราะห์ Phylogenetic analysis พบว่า ผลที่ได้เป็น subgenotype 1A และ จากการทำ Phylogenetic analysis ของทั้งจีโนมของไวรัสในตัวอย่างซีรัมของจ.ลำปาง (LP014) พบว่าจัดอยู่ใน subgenotype 1A จากผลการทดลองแสดงว่า subgenotype 1A เป็น subgenotype หลักที่หมุนเวียนในประเทศไทย.	en
dc.description.abstractalternative	During 2001-2005, many outbreaks of hepatitis A virus (HAV) infection have occurred in Thailand. We studied the HAV genotypes, performed phylogenetic analysis on various HAV isolates obtained from the different outbreaks during 2001-2005 in Thailand and finally, examined the entire genome of one selected HAV isolate. Serum samples obtained from four epidemic provinces in Thailand including Suphanburi (N = 83), Songkhla (N = 80), Chiangrai (N = 61) and Lampang (N = 59) were tested for anti-HAV IgM by ELISA. Anti-HAV IgM was detected in 40 (48.19%), 38 (47.5%), 25 (40.98%) and 32 (54.24%) samples from Suphanburi, Songkhla, Chiangrai and Lampang, respectively. HAV-RNA was reverse transcribed and amplified by nested PCR focused on the VP1-2A region. The VP1-2A region of all HAV-RNA positive samples was subjected to nucleotide sequencing. Molecular characterization and phylogenetic analysis revealed that all isolates from these outbreaks clustered in subgenotype IA and hence, are closely related to isolates previously investigated in Thailand. One sample from Lampang (LP014) was selected for entire genome sequencing. Resulting genome comparison and phylogenetic analysis showed that LP014 also clusterd in subgenotype 1A. The presence of a single sub-genotype indicates that subgenotype 1A has been the one predominantly circulating strain in Thailand.	en
