dc.description.abstract	จุดประสงค์ของงานวิจัยนี้ คือ ศึกษาการตอบสนองของเซลล์เอ็นยึดปริทันต์ เมื่อกระตุ้นด้วยเอสโตรเจน IL-1beta และ LPS โดยลำพัง หรือเมื่อกระตุ้นด้วยเอสโตรเจนร่วมกับ IL-1beta หรือเอสโตรเจนร่วมกับ LPS โดยต้องการตรวจสอบการตอบสนองในแง่การแสดงออกของ IL-6, NOS และการหลั่งเอนไซม์ MMP-1, -2,-3 เซลล์เอ็นยึดปริทันต์ถูกเพาะเลี้ยงจากเนื้อเยื่อเอ็นยึดปริทันต์ของมนุษย์ เซลล์ถูกกระตุ้นด้วยสารกระตุ้น เป็นเวลา 24 ชั่วโมง และตรวจสอบการเปลี่ยนแปลงระดับการแสดงออกของ IL-6 และ NOS ด้วยเทคนิคอาร์ที-พีซีอาร์ ส่วนการเปลี่ยนแปลงของระดับเอนไซม์ MMP-1 และ MMP-2, -3 จะตรวจสอบด้วยวิธีเวสเทอร์นอนาไลซิส และไซโมกราฟีตามลำดับ หลังจากกระตุ้นเซลล์เป็นเวลา 48 ชั่วโมงผลการทดลองพบว่า เอสโตรเจนที่ความเข้มข้น 100 นาโนโมลาร์ มีผลกระตุ้นการหลั่ง MMP-3 แต่ไม่มีผลต่อ MMP-1,-2 และไม่มีผลต่อการแสดงออกของ IL-6 และ NOS ในขณะที่ IL-1 beta และ LPS มีผลกระตุ้นการแสดงออกของ IL-6 และ NOS และเพิ่มการหลั่ง MMP-1 ประเด็นที่น่าสนใจ คือ เอสโตรเจนช่วยเสริมผลของ IL-1 beta ในการเหนี่ยวนำการแสดงออกของ IL-6, NOS และ MMP-1 โดยจะทำให้ระดับของ IL-6, NOS และ MMP-1 เพิ่มขึ้นเป็น 2.4, 5 และ 2.3 เท่า ตามลำดับ เมื่อเทียบกับ 1.7, 4.2 และ 1.3 เท่า เมื่อกระตุ้นด้วย IL-1beta เพียงลำพัง ในขณะเดียวกันเอสโตรเจนยังช่วยเสริมผลของ LPS ด้วย โดยระดับของ NOS และ MMP-1 เพิ่มขึ้นเป็น 3.7 และ 2.9 เท่าตามลำดับเมื่อเทียบกับ 2 และ 1.9 เท่า เมื่อกระตุ้นด้วย LPS ตามลำพัง ผลการทดลองนี้ แสดงให้เห็นว่า ในสภาวะที่มีระดับของฮอร์โมนเอสโตรเจนเพิ่มสูงขึ้น จะมีผลต่อการตอบสนองของเซลล์เอ็นยึดปริทันต์ที่มีต่อสารกระตุ้นที่ก่อให้เกิดการอักเสบ โดยทำให้ระดับของเอนไซม์และซัยโตไคน์ที่เกี่ยวข้องกับการทำลายเนื้อเยื่อเพิ่มสูงขึ้น ซึ่งอาจเป็นสาเหตุหนึ่งที่ทำให้ระดับของการอักเสบของเนื้อเยื่อปริทันต์เพิ่มสูงขึ้นในระยะวัยรุ่น หรือในสตรีที่มีครรภ์ ซึ่งมีระดับของฮอร์โมนเอสโตรเจนเพิ่มสูงขึ้นในกระแสเลือด	en
dc.description.abstractalternative	The purpose of this study is to investigate the response of periodontal ligament cells after activating with estrogen, IL-1beta or lipopolysaccharides (LPS) alone or activating with the combination of estrogen and IL-1beta or estrogen and LPS. After 24 hours of the activation the levels of IL-6 and NOS expressions were examined using RT-PCR analysis. The amounts of MMP-1, -2 and -3 in the medium were also measured after 48 hours activation using Western analysis for MMP- 1 and zymography for MMP-2 and MMP-3. The results indicated that estrogen alone had no effect on IL-6, NOS, MMP-1 and MMP-2 but could increase MMP-3 expression and also increased the secretion of MMP-1. Interestingly, when cells were activated with both estrogen and IL-1beta, the levels of IL-6, NOS and MMP-1 increased up to2.4, 5 and 2.3 folds, respectively, compared with 1.7, 4.2 and 1.3 folds when cells received IL-1beta alone. Similarity, activation with both estrogen and LPS increased the level of NOS and MMP-1 from 2 and 1.9 fold up to 3.7 and 2.9 folds, respectively, in comparing with LPS-treated alone. The results showed that increasing level of estrogen, especially during puberty and pregnancy, could affect the response of PDL cells to pro-inflammatory factors by increasing the enzyme or cytokine release, which consequently enhanced periodontal tissue destruction.	en
