| Abstract: | การวัดปริมาณของ D-norgestre1 ด้วยวิธี radioimmunoassay ในรายงานนี้ เสนอผลการพัฒนาวิธี การหาปริมาณของ D-norgestre1 โดยใช้สารติดสลากรังสีคือ D-norgestre1-3-oxime/(¹²⁵L)-iodohistamine แอนตีบอดีที่ใช้ได้จากการฉีดกระต่ายด้วย D-norgestre1-3-oxime-BSA จากการศึกษาหาสภาวะต่าง ๆ ที่เหมาะสมสำหรับปฏิกริยา radioimmunoassay ของ D-norgestre1 พบว่า ความเข้มข้นสุดท้ายที่เหมาะสมของแอนดีบอดีเป็น 1 : 160,000 และปริมาณของอนุพันธ์ที่ติดสลากเป็น 5,000 - 10,000 cpm/tube เวลาและอุณหภูมิที่ได้ incubate ประมาณ 18 – 24 ชั่วโมง ที่ 4 ℃ การแยก D-norgestre1 รูปอิสระออกจากรูปที่วัยกับแอนตีบอดีใช้วิธีแยกด้วยผงถ่านปริมาณ 2.5 mg/tube ใน phosphate buffer PH 7.4 ที่อุณหภูมิ 4 ℃ ปฏิกิริยาการดูดซับของผงถ่านไม่แตกต่างไปจากเดิมเมื่อเติมและไม่เติม 100 /ul ของ 0.5% gelatin phosphate buffer ส่วน dextran T 70 ปริมาณตั้งแต่ 0.05 – 3.2 mg/tube พบว่ามีผลต่อการดูดซับของผงถ่าน แต่ไม่ทำให้ลักษณะของกราฟมาตรฐานเปลี่ยนไป การหาปริมาณของ D-norgestre1 ในน้ำนมตัวอย่างทำโดยใช้วิธี คัดด้วย ether แล้วกำจัดสารเจือปน โดยใช้ sephadex LH 20 column และชะด้วย isooctane : benzene : methanol 70 : 20 : 10 โดยปริมาตร ส่วนการหาปริมาณของสารนี้ ในซีรัมนั้นใช้สกัดโดยวิธีเดียวกับน้ำนมแต่ไม่จำเป็นต้องผ่าน sephadex column จากการทดสอบความเชื่อถือได้ของวิธีทดลองนี้ พบว่า ความแม่นยำในการทำปฏิกริยาเดียวกันและในระหว่างการทำปฏิกิริยาทั้งในน้ำนมและซีรัม มีสัมประสิทธิ์ของการเปลี่ยนแปรน้อยกว่า 10% ความถูกต้องของการวัดจะเปลี่ยนแปลงได้บ้าง เมื่อปริมาณของ D-norgestre1 ในสารตัวอย่างเปลี่ยนไป ดังนั้น ในการ วัดปริมาณจากสารตัวอย่างทั้งในน้ำนมและซีรัมนั้น ได้เลือกได้ปริมาณของสารตัวอย่างให้อยู่ในช่วงซึ่งความ ถูกต้องอยู่ระหว่าง 90 ± 10% ทุกครั้ง ความไวของการทดลองเป็น 5 pg/cm³ แอนตีบอดีที่ใช้มีความจำ เพาะสูง กล่าวคือ มีปฏกริยาไม่เกิน 30% กับ metabolite บางชนิดของ D-norgestre1 เท่านั้น ผลการวัดปริมาณของสารนี้ในน้ำนมและซีรัมของสตรีอาสาสมัคร 6 ราย ที่กินยาคุมกำเนิด Microlut ซึ่งประกอบด้วย D-norgestre1 30 ug /วัน ปรากฏว่า ปริมาณของสารที่วัดได้อยู่ระหว่าง 20 – 160 pg/cm³ และ 0.2 – 1.5 ng/cm³ ของน้ำนมซีรัมตามลำดับ |
| Other Abstract: | A radioimmunoassay of D-norgestre1-3-oxime/(¹²⁵I)-iodohistaminc as radioactive ligand has been developed. The antibody used was raised in rabbit against D-norgestre1 -3-oxime-BSA and a final dilution of 1 : 160,000 was found to be most practical and highly Satisfactory. No significant different binding was obtained with the radio higand ranging from 5,000 -10,000 cpm/tube. The incubation was performed at 4 ℃ for 18 – 24 hour, and the Separation of free and bound form was accomplished by the addition of 2.5 mg charcoal in phosphate buffer PH 7.4 at 4 ℃ The adsorption of charcoal was not significantly altered by 0.5% gelatin phosphate buffer , but altered by 0.05 – 3.2 mg/tube of dextran T 70, but these concentration cannot interfere with the shape of standard curve of D-norgestre1. An ether extract of milk samples were purified by sephadex LH 20 column and eluted with isooctane : benzene : methanol 70 : 20 : 10 V/V before assay. The determination of D-norgestre1 in Serum was satisfactory without the chromatographic step. The reliability of the method showed that coefficient of variation was less than 10% both for within and between assay. Since the accuracy of the assay varied with the concentration of the D-norgestre1 in the reaction media, the final assay of the sample was carried out by adjusting the concentration to the level which the accuracy range fall within 90 ± 10%. A sensitivity of 5 pg/cm³ was obtained. The antibody was considered to highly spccific showing cross reaction of not more than 30% with only some of D-norgestre1 derivatives. Determination of this synthetic steroid in milk and serum from 6 subjects who used pill containing D-norgestre1 30 µg/day showed significant amounts ranging from 20 - 160 pg/cm³ in milk and 0.2 – 1.5 ng/cm³ in serum. |
