| Abstract: | เพื่อศึกษาความเหมาะสมของน้ำเพาะเลี้ยงในการคัดเลือกตัวอสุจิและความสามารถในการเจาะทะลุไข่แฮมสเตอร์ของตัวอสุจิคน โดยใช้น้ำเพาะเลี้ยงชนิดต่าง ๆ 3 ชนิด (Ham's F-10, BWW และ TMPA) ที่มีความเข้มข้นของ Human Preovulatory Serum (HPS) ร้อยละ 20 พบว่าตัวอสุจิเคลื่อนไหวได้ดีและมีอัตราการเจาะทะลุไข่แฮมสเตอร์สูงในน้ำเพาะเลี้ยง BWW + 20% HPS มากกว่าในน้ำเพาะเลี้ยงชนิดอื่น จึงเลือกใช้น้ำเพาะเลี้ยง BWW +20% HPS ในการศึกษาความสัมพันธ์ระหว่างลักษณะต่าง ๆ ที่พบจากการวิเคราะห์น้ำอสุจิกับการเจาะทะลุไข่แฮมสเตอร์ที่ไม่มีโซนา ผลการวิจัยพบว่า ค่าเฉลี่ยของอัตราการเจาะทะลุไข่แฮมสเตอร์ของตัวอสุจิที่ได้จากชายที่ภรรยากำลังตั้งครรภ์ภายใน 12 สัปดาห์ (กลุ่มควบคุม) (40.63±12.59) มีค่าสูงกว่าของตัวอสุจิที่ได้จากชายที่มีบุตรยาก (กลุ่มศึกษา) (6.43±8.90 ) อย่างมีนัยสำคัญทางสถิติ (P<0.001) และพบว่า มีความสัมพันธ์กันอย่างมีนัยสำคัญที่ระดับ .05 ระหว่างอัตราการเจาะทะลุไข่เฮมสเตอร์กับ 3 ค่าพารามิเตอร์ของการวิเคราะห์น้ำอสุจิ : เปอร์เซ็นต์ของการเคลื่อนไหว (r=0.588), เปอร์เซ็นต์ตัวอสุจิเป็น (r =0.509) และความเข้มข้นของตัวอสุจิ (r =0.499) แต่ความสัมพันธ์เหล่านี้ค่อนข้างน้อยมาก |
| Other Abstract: | Three different culture media (Han's F-10, BWW and TMPA) supplemented with 20% Human Preovulatory Serum (HPS) were tested for their suitability in sperm-washed procedure and penetrating ability of human spermatozoa. It was found that BWW + 20% HPS yielded higher percentage of sperm motility and penetration rates than the other. BWW medium was then used for the study of the correlation between the different parameters of routine semen analysis and the zona-free hamster egg penetration assay. It was found that the mean penetration rate of sperm from men whose wives were pregnant during 12 weeks (control group) (40.63±12.59) was significantly higher (p<0.001) than that of the voluntary men from the infertile clinic (study group) (6.43t8.09). The correlation (p<0.05) between the penetration rate and three parameters of the semen analysis: the percentage of sperm motility, the percentage of viability and the concentration were r=0.588, 0.509 and 0.499 respectively. |
